Your initial coarse grain screen is a vital first step in crystallisation. The SG1 sparse matrix kit from Janet Newman (CSIRO) gives you the most successful conditions from the most popular commercial screens in one kit. Now we're pairing it with the Ligand-Friendly Screen, the most successful grid screen in regular use at the SGC, in our NextGen Combo kit to give your crystallisation the best start available.

This is the ECO version of the kit in which Cacodylate is replaced by MES.

The SG1 Screen
The term 'shotgun screening' was coined early in the Structural Genomics era and refers to the process of setting up experiments using pre-mixed cocktails until a crystal of sufficient quality is obtained. The SG1™ Screen uses the ShotGun approach to gather all the most successful conditions from all the early commercially available screens.
Developed by Prof Janet Newman at CSIRO, the C3 ShotGun screen (SG1™) is a set of 96 crystallisation conditions that occur with highest, non-redundant frequency amongst structures deposited in the PDB.

The Ligand-Friendly Screen (LFS)1 has been one of the two primary coarse screens used at the Structural Genomics Consortium (SGC) in Oxford since 2004; at last count, it had produced 190 novel structures over 10 years.
  • Identify more tight-binding ligands, as it contains no components that are likely to bind at protein-ligand interaction sites.
  • Near physiological pH conditions ensure you are studying the active form of your protein.
  • More hits for rarely-crystallising proteins. The semi-systematic format of the screen provides multiple trials of similar conditions - an important factor for difficult cases.
  • Increased hit rate for ligand co-crystallisations – the semi-systematic format efficiently screens around related conditions when complex formation alters crystallisation conditions.
  • In use at the SGC for more than 10 years, crystals grown in the screen conditions have resulted in 190 structures so far, including those of Haspin2 and CLK33 in recent publications.
